<正> 魚精蛋白的分子量较小,所含氨基酸成分也较简单,它与核酸结合存在于成熟的魚精子头部。魚精蛋白之间有种属特異性的差異,它们的N—末端氨基酸亦不相同,例如日本北海道产的鯡魚(Clupea pallasij)精蛋白的N—末端是丙氨酸,而欧洲产的鲱魚(Clupea havengus)精蛋白的N—末端则是脯氨酸。关于中国沿海各种魚类的魚精蛋白的研究不多,因此研究魚精蛋白的成分、结构、与核酸结合的关系、本身形成的过程以及其生物学的功能等,是很有意义的。我们用纸上层析法,对我国黄海南部所产的河触和鮁焦精蛋白所含的氨基酸作了初步测定。
Abstract:The present paper is a preliminary report on the amino acid composition of the protamines of Spheroides yermicularis radiata Abe and Sawara niphonia Cuvier & Valenciennes, determined with paper chromatography. The isolation and purification of the protamines was made mainly by Ando's method. The purified protamines obtained were white powder-like.The hydrolysates of the purified protamines were analized with one-dimensional ascending paper chromatography. The temperature was controlled at 37°±0.5℃. The solvents used were : I. n-butyl alcohol: water; acetic acid = 4: 5: Ⅰ; Ⅱ. n-amyl alcohol : pyridine: water =35: 35: 30; Ⅲ. phenol: water = 70:30. Ninhydrin and isatin were used as developing agents. It was found that the amino acid composition of the protamine3 and their Rf values ( for solvent I ) were as follows:Both protamines were free of cyclic, dicar boxylic and sulfurconta-ining amino acids. Judged from the colored .spots, the isoleucine and threonine contents in the protamine of S. niphonia appeared to be less than those in the protamine of Sph. vermicularis, and the arginine content. higher.
