外源脱落酸对鳗草种子休眠的延长作用

摘要:

为探究脱落酸(Abscisic acid, ABA)对鳗草种子休眠的影响,本研究设置了0(对照)、3、15、30和300 mg·L-1 ABA浓度处理组,通过监测种子的萌发率、种子活力、再萌发率,探究了外源ABA对延长鳗草(Zostera marina)种子休眠的作用,分析了鳗草种子应对外源ABA的生理响应过程。研究发现:(1)外源ABA的添加对鳗草种子休眠具有显著影响,其中30 mg·L-1处理组种子萌发率、活力和再萌发率分别为对照组的41%、1.4倍和2.8倍。(2)受外源ABA的影响,延长休眠期内,高浓度处理组(30和300 mg·L-1)种子内部可溶性糖含量呈下降趋势,淀粉和可溶性蛋白含量呈上升趋势,水分含量上升不明显。至实验结束,30 mg·L-1处理组种子内部的可溶性糖和水分含量最低,分别为对照组的78%和88%,淀粉和可溶性蛋白含量最高,为对照组的1.2和1.6倍。(3)相关性分析显示,水分含量是影响种子休眠的重要物质,其与萌发率呈极显著正相关,与种子活力、再萌发率呈极显著负相关。综合分析认为,30 mg·L-1的外源ABA是延长鳗草种子休眠的适宜抑制剂,其主要通过降低种子的吸涨作用实现种子休眠的延长。研究结果可为鳗草种质资源储存和规模化增殖提供理论依据和科学基础。

Abstract:

To investigate the effect of Abscisic acid(ABA) on the seed dormancy of eelgrass, five different treatment groups(0, the control, 3, 15, 30 and 300 mg·L-1 ABA) were set in this study. The effect of ABA on the seed dormancy of eelgrass was investigated by monitoring the germination rate, seed vigor and regermination rate during the prolonged dormancy period. Physiological responses of eelgrass seeds to the exogenous ABA were investigated by monitoring the changes of stored nutrient substances in seeds during the prolonged dormancy period. The results showed that exogenous ABA addition effectively prolonged the seed dormancy. The concentration of 30 mg·L-1 ABA displayed the best prolongation effort, 41% lower than germination rate of control, and 1.4 times and 2.8 times higher than the vigor and regermination rate of seeds of control. The soluble sugar content of seeds in higher concentration groups(30 and 300 mg·L-1) showed a decreasing trend during the prolonged dormancy period while the content of water, starch and soluble protein of seeds exhibited an increasing trend. At the end of the experiment, the content of soluble sugar and water of seeds in 30 mg·L-1 treatment group attained the lowest values which were 78% and 88% lower than those of seeds of control, respectively. The contents of starch and soluble protein of seeds attained the highest values which were 1.2 and 1.6 times higher than those of seeds of control, respectively. Correlation analysis of physiological and ecological indexes showed that water content of seeds was important; it affects the seed dormancy and significantly and positively correlates with the seed germination rate and significantly and negatively with seed vigor and regermination rate. Comprehensive analysis showed that exogenous ABA addition effectively prolonged the seed dormancy, and 30 mg·L-1 of ABA was optimal for prolonging seed dormancy through reducing the water absorption of seeds. These results provided a theoretical and scientific basis for the storage of germplasm resources and the large scale enhancement of eelgrass.
