凡纳滨对虾组织中虾青素的分析方法建立、分布特征及功能探究

摘要:

为探究虾青素在凡纳滨对虾(Litopenaeus vannamei)体内的分布特征,本研究建立了一种虾青素提取与定量分析方法,确定甲醇为最佳萃取溶剂,采用1∶50固液比单次萃取,结合“甲醇萃取-四氢呋喃复溶”联用检测方案。饲料中虾青素添加量分别为0(对照组)、40、80、120 mg/kg,进行投喂实验,探究不同添加量虾青素对凡纳滨对虾的影响,并定量分析虾青素分布、体色变化及酶活性。研究表明,虾青素能够改善对虾体色,使其向红橙色转变,并在肝胰腺、腹神经索和表皮中积累最为显著。此外,虾青素能够提高肝胰腺总抗氧化能力(Total antioxidant capacity, T-AOC),增强超氧化物歧化酶(Superoxide dismutase, SOD)、过氧化氢酶(Catalase, CAT)、碱性磷酸酶(Alkaline phosphatase, AKP)和谷胱甘肽过氧化物酶(Glutathione peroxidase, GSH-Px)活性,同时降低丙二醛(Malondialdehyde, MDA)含量,表明虾青素可有效增强凡纳滨对虾的抗氧化与免疫功能。

Abstract:

To elucidate the distribution characteristics of astaxanthin in Pacific white shrimp(Litopenaeus vannamei), a robust method for the extraction and quantification of astaxanthin in shrimp tissues was developed. Methanol was identified as the optimal extraction solvent. A single-step extraction at a 1∶50(w/v) solid-liquid ratio combined with a methanol-THF sequential dissolution approach was utilized. Through controlled feeding trials with graded astaxanthin supplementation(0, 40, 80, 120 mg/kg), we quantitatively analyzed the tissue-specific accumulation patterns, body color responses and associated enzymatic changes. The results indicated that astaxanthin significantly enhanced the body color of shrimp to a red-orange hue and was predominantly accumulated in the hepatopancreas, ventral nerve cord and epidermis. Additionally, astaxanthin supplementation effectively increased the total antioxidant capacity(Total antioxidant capacity, T-AOC) of the hepatopancreas, elevated the activities of superoxide dismutase(Superoxide dismutase, SOD), catalase(Catalase, CAT), alkaline phosphatase(Alkaline phosphatase, AKP) and glutathione peroxidase(Glutathione peroxidase, GSH-Px), and reduced the content of malondialdehyde(Malondialdehyde, MDA).These findings demonstrated that astaxanthin significantly enhances the antioxidant capacity and immune function of L. vannamei.
