cd36基因在斑马鱼抗鳗弧菌免疫调控中的作用The Role of cd36 Gene in Immune Regulation Against Vibrio anguillarum in Zebrafish

摘要(Abstract):
为了研究清道夫受体CD36在鱼类抗细菌免疫调控中的作用,本文将鳗弧菌(Vibrio anguillarum)腹腔注射到cd36基因敲除的斑马鱼(cd36~(-/-))和野生型斑马鱼(WT)中,发现突变体斑马鱼的活跃度和存活率均低于野生斑马鱼。分别取鳗弧菌(V.anguillarum)感染的cd36~(-/-)和WT斑马鱼头肾组织进行转录组学测序分析,发现二者差异表达的基因主要富集在补体系统(Complement system)、系统性红斑狼疮(Systemic lupus erythematosus)、细胞因子之间的相互作用(Interaction between cytokines)和病毒致癌机理(Viral carcinogenesis)等与免疫相关的信号途径中。运用实时荧光定量PCR技术进一步证实,与野生型斑马鱼相比,cd36~(-/-)斑马鱼中参与抗原呈递的mhc1和cd8基因、穿孔素基因、补体系统关键基因c3以及细胞因子基因il-4的表达都显著下调。综上,本研究证实CD36在斑马鱼抗鳗弧菌免疫调控中发挥重要作用。
