苯酚-硫酸法测定黄河口沉积物中的多糖Phenol-Sulfuric Acid Method for the Analysis of Polysaccharides in Sediment at the Yellow River Estuary

摘要(Abstract):
建立苯酚-硫酸法测定黄河口沉积物中的多糖,确定了最佳实验条件:最大吸收波长为487nm,苯酚质量分数为12%,95.5%的浓硫酸用量为4mL,溶液反应体积为2.5mL,绘制标准曲线所采用的标准物质为含有0.1g沉积物空白样品和一定量葡萄糖的混合物。结果表明,在优化实验条件下,成功获得多糖与葡萄糖之间的换算因子为4.85。该方法的精密度为4.4%,平均回收率为98.68%,相对标准偏差为2.3%。应用本方法测出黄河口沉积物中的多糖平均含量为0.1%,相对标准偏差为2.8%,与有关文献报道相符合,表明该方法能够满足黄河口沉积物中多糖含量测定的要求。
